【Objective】To study the effects of shRNA-mediated CIP2A gene silencing on the proliferation,apoptosis and invasion of human colorectal cancer HT29 cells.【Methods】The shRNA-CIP2A was transfected into human colorectal cancer HT29 cells. The expression levels of CIP2A mRNA and protein were determined by real-time PCR and Westernblot,respectively. Cell proliferation,apoptosis and invasion were determined by transwell assay,flow cytometry (FCM),tumor implantations experiment(n=20)and CCK8,respectively.【Results】The mRNA and protein expression of CIP2A in human colorectal cancer HT29 cells was reduced in HT29 cells transfected with shRNA targeting CIP2A gene. The HT29 cells transfected with shRNA targeting CIP2A gene showed a significant decrease of cell proliferation, clonogenicability,invasivenes sand tumorigenicity(453.0±7.5vs152.0±4.7,n=3,P=0.001)when compared with the control. The apoptoticrate(%)of control group was lower than in the CIP2A shRNA group(11.09±0.32vs14.25± 0.04,n=3,P=0.001) .【Conclusion】CIP2A knock down led to substantial reductions of proliferation and invasion of HT29 cells and induced their apoptosis,indicating that CIP2A may be a potential target for colorectal cancer therapy.
