【目的】 寻求一种安全、微创、可信的构建椎动脉型颈椎病动物模型的方法。【方法】 高频超声监测正常猕猴(6只,雌雄各半,年龄3.5 ~ 5.5岁,体质量4 ~ 6 kg)的椎动脉血流状况,在高频超声的引导下通过骨穿针在猕猴颈C5-6左侧椎动脉旁注射复合重组人骨形态发生蛋白-2 (rhBMP-2 )的磷酸钙骨水泥1.5 mL来制作椎动脉型颈椎病模型,并用高频超声监测造模后椎动脉收缩期峰值血流(VS) 、舒张期末峰值血流(VD)、阻力指数(RI)、血管内径(D)并评价猕猴椎动脉血流的改变是否符合临床上椎动脉型颈椎病患者血流的改变。【结果】 发现在造模2个月后猕猴椎动脉的血流变化较明显,且趋于稳定,造模2月后左侧椎动脉血流速度减慢,阻力指数增高,血管管径变窄,VS(6.30 ± 1.52)cm/s、VD(2.41 ± 0.66)cm/s、RI(0.71 ± 0.04)、D(0.08 ± 0.01)cm,同造模前比较,差异均具有统计学意义(P < 0.05)。右侧椎动脉代偿性的血流速增加,管径增粗,RI减低,VS(8.87 ± 0.98)cm/s、VD(4.31 ± 0.12)cm/s、RI(0.57 ± 0.41)、D(0.09 ± 0.01)cm,同造模前比较P > 0.05,差异均无统计学意义。【结论】 高频超声引导注射骨水泥制作猕猴椎动脉型颈椎病模型是一种安全、可行的实验方法。
【Objective】 In order to build a safe, minimally invasive and high credibility animal model of vertebral artery cervical spondylopathy. 【Methods】 six health adults macaque monkeys aged 3.5 ~ 5.5 years and weighing from 4 to 6 kg, were selected as candidates for this study. High -resolution ultrasound monitor the blood flow parameters of vertebral artery of macaque monkey, high -resolution ultrasound -guide injected injectable 1.5mL fast -setting calcium phosphate cement (CPC) with rhBMP -2 to pressed or stimulated the left vertebral artery between C5 and C6 of macaque monkey. We examined and recorded systolic peak flow velocity (VS), diastolic peak flow velocity (VD), resistance index (RI) and vascular diameter (D) of vertebral artery after the injected, and use high -resolution ultrasound to evaluate the effect of the experimental model. 【Results】 we found that after two months later the blood flow parameters of vertebral artery of macaque monkey approximately trends to be stable, after the modeling the left side of the vertebral artery course bend, vascular inner of the left side became narrow and blood flow velocity decreased, VS of left side of the vertebral artery was (6.30 ± 1.52)cm/s, VD of left side of the vertebral artery was(2.41 ± 0.66)cm/s, RI of left side of the vertebral artery was(0.71± 0.04), D of left side of the vertebral artery was(0.08 ± 0.01)cm, before and after the building the blood flow parameters of the left vertebral artery of macaque monkey have a significant difference (P < 0.05), with statistical significance; vascular inner and blood flow velocity of the right side compensatory increase, RI decrease, VS of right side of the vertebral artery was (8.87 ± 0.98)cm/s, VD of right side of the vertebral artery was(4.31 ± 0.12)cm/s, RI of right side of the vertebral artery was(0.57 ± 0.41), D of right side of the vertebral artery was(0.09 ± 0.01)cm, However, before and after the building the blood flow parameters of the right vertebral artery of macaque monkey have no significant difference (P > 0.05), without statistical significance. 【Conclusion】 The experimental method by injecting rhBMP -2 bone cement to press or stimulated the left vertebral artery of macaque monkey to make cervical vertebral disease was a reliable and safe.
