【Objective】To observe the effect of microRNA16 on the biological behaviors and the expression of Wip1 of the human glioblastoma U87 cell.【Methods】The cell model of miR16 upregulated was built. Cellular proliferation was assayed by the colony formation assay. The apoptosis capacity and the cell cycle were assayed by flow cytometry. Cellular migration ability was detected by transwell chamber assay. The expression of miR16 and Wip1 mRNA were measured by qRT-PCR
and Western blotting was used to measure the protein expression of Wip1 and p53. 【Results】QRT- PCR showed that the miR16 expression transfected with mimic group had been enhanced significantly. The rates of cell proliferation in miR16 mimic group and NC group were (7.64±0.61)% and (21.47±0.61)%
respectively; the colony forming rates were (19.17±0.76)% and (33.83±3.75)% respectively; the early apoptosis rates were (5.01±0.64)% and (2.37±0.82)%
respectively; the cell proportions in S phase were (23.66±0.39)% and (28.98±0.08)%
respectively
and the average number of migration cells under 200 magnification field were (74.33±2.52) and (137.67±9.61)
respectively. MiR16 over-expression significantly inhibited U87 cell proliferation and invasion
also it promoted U87 cell apoptosis
and induced Wip1 down expression and P53 up expression.【Conclusion】MiR16 suppressedU87 cells proliferation and invasion by targeting Wip1.
