摘 要: 【目的】 观察人脐带间充质干细胞(hUC-MSC)移植对小鼠慢性输卵管炎的治疗作用,为慢性输卵管炎的临床治疗提供新策略。【方法】 从脐带中分离培养人脐带间充质干细胞,流式细胞术鉴定其免疫表型,并诱导成脂、成骨分化验证其多向分化潜能。通过阴道内接种沙眼衣原体建立小鼠输卵管炎模型,感染后4周随机分成hUC-MSC移植组及对照组。hUC-MSC移植组小鼠阴道内接种人脐带间充质干细胞,对照组小鼠阴道内注射等体积PBS。感染后8周,观察小鼠输卵管有无阻塞、积水等慢性输卵管炎表现,病理切片观察组织形态学改变。【结果】 分离培养的人脐带间充质干细胞符合间充质干细胞的一般生物学特性。感染后8周,对照组小鼠均出现输卵管阻塞、积水等慢性输卵管炎表现,而hUC-MSC移植组仅2例出现输卵管阻塞及积水,对照组小鼠慢性输卵管炎的发生率明显高于hUC-MSC移植组,差异有统计学意义(P< 0.01)。hUC-MSC移植组输卵管病理切片显示,输卵管周围炎症细胞浸润较少。【结论】 hUC-MSC移植可明显减少小鼠慢性输卵管炎的发生和减轻其炎症程度,有可能为输卵管性不孕的临床治疗提供新的途径。
Abstract: 【Objective】 To investigate the effect of human umbilical cord mesenchymalstem cells (hUCMSC) transplantation treatment on mice chronic salpingitis, and toprovide new tactics for clinical therapy of chronic salpingitis. 【Methods】 hUCMSCswere isolated from human umbilical cord and cultured in vitro. The phenotypic characteristicsof hUCMSC were determined by FACS and multilineage differentiation capacity wasconfirmed by induced adipogenesis and osteogenasis. The salpingitis models of micewere established by intravaginal inoculation of the Chlamydia trachomatis. All micewere randomly divided into hUCMSC transplantation group and control group at 4 weeksafter infection. Mice of hUCMSC transplantation group were transplanted by hUCMSCsthrough intravaginal inoculation, and control group were injected by PBS. At 8 weeksafter infection, all mice were sacrificed to evaluate the status of oviduct occlussionor hydrosalpinx. Besides, histopathological changes of oviduct were examined bylight microscopy. 【Results】 hUCMSC cultured in vitro exhibited general biologicalproperties of mesenchymal stem cells. At 8 weeks after infection, the incidenceof salpingemphraxis and hydrosalpinx in control group was obviously higher thanthat of hUCMSC transplantation group, and the difference was of statistical significance(P < 0.01). Moreover, tubal pathology section showed that inflammatory cells infiltrationwas less in hUCMSC transplantation group than that of control group. 【Conclusion】hUCMSC transplantation could reduce the occurrence of mice chronic salpingitis and improve tubalinflammation, which may serve as a new methodfor the clinical treatment of tubal infertility.
