摘 要: 【目的】 观察长链非编码RNA(lncRNA)在正常舌组织和舌鳞癌组织中表达谱的变化。 【方法】 利用lncRNA表达谱芯片检测技术,筛查3例舌鳞癌组织及3例正常舌组织中lncRNA表达谱的变异,通过对原始数据进行预处理达到均一化后,确定舌鳞癌组织中2倍以上变化并有显著差异(P<0.05 ) 的lncRNA为差异表达lncRNA,筛选出差异表达的lncRNA;分别在scc-9、cal-27、um-1和um-2共4种人舌鳞癌细胞和1种人正常舌细胞以及在35例舌鳞癌组织和12例正常舌组织中,利用实时荧光定量逆转录-多聚酶链反应(qRT-PCR)对芯片结果进行验证;对筛选所得的部分差异表达的lncRNA,利用lncRNA与mRNA特异性表达的标准化信号强度,构建基因共表达网络,并进行分析。【结果】 芯片共筛选差异表达lncRNA 3590条,其中上调的共1785条,降低的共 1805条;选取其中部分lncRNA,qRT-PCR进一步证实了芯片结果的准确性;共表达网络构建提示某些lncRNA可能在参与舌鳞癌发生发展过程中发挥重要调控作用。【结论】与正常舌组织比较,舌鳞癌组织lncRNA表达谱发生显著变化,提示差异表达的lncRNA 可能与舌鳞癌的多种恶性表型有着密切联系。
Abstract: 【Objective】 To analyze the expression profile variation of long non-coding RNA (lncRNA) in tongue squamous cell carcinoma (TSCC)and normal tongue tissue (NTT). 【Methods】 To use lncRNA microarray technology toinspect the difference of lncRNA expression in 3 cases of TSCC tissues and 3 cases of normal tonguetissues, and then make analysis after pretreatment and homogenization of raw data,to identify which have more than 2 times variation and significant difference (P< 0.05) by statistical analysis as lncRNA with differential expression. The lncRNAexpression was confirmed by quantitative real-time polymerasechain reaction (qRT-PCR) in four kinds of human tongue squamouscell carcinoma cell lines (scc-9, cal-27,um-1,um-2) and 1 case of normaltongue cell line, but also in 35 cases of TSCC tissues and 12 cases of normal tongue tissues.Gene co-expression network was constructed according tothe normalized signal intensity of specific expression lncRNA and mRNA. 【Results】Compared with NTT, there are 3590 lncRNA differential expressed, while 1785 increasemore than 2 times and 1805 reduce more than 2 times. Some of these lncRNA were chosen,the accuracy of the results was verified by qRT-PCR. Geneco-expression network prompted that some lncRNA may playan important role in the process of in TSCC. 【Conclusion】 lncRNA expression profileof the TSCC changes significantly in comparison with the NTT, the different lncRNAmay have a close connection with many kinds of malignant phenotype in the TSCC.
