摘 要: 【目的】 分析早期鼻咽癌的基因甲基化状态
初步构建早期鼻咽癌的基因甲基化谱?【方法】 收集I期(T1N0M0)鼻咽癌石蜡标本及炎性增生性鼻咽组织各4例
分别提取DNA
富集两组样品基因组甲基化片段后采用甲基化芯片(NimbleGen)对两组标本进行高通量测序分析
对杂交信号进行扫描及初步数据处理
分析并筛选出两组标本的差异甲基化基因?【结果】 4例鼻咽癌石蜡标本中均出现甲基化的基因数目为1 134个
而4例炎性增生性鼻咽组织中均出现甲基化的基因有2 091个
4例鼻咽癌标本全部发生甲基化但4例炎性增生性鼻咽组织全未发生甲基化的基因有218个
提示此218个基因可能为早期鼻咽癌患者的甲基化基因谱?另外
4例鼻咽癌标本全未发生甲基化而4例炎性增生性鼻咽组织全部发生甲基化的基因数目为1 558个?【结论】 本研究初步构建了早期鼻咽癌的基因甲基化谱
为进一步建立更为精确的甲基化谱奠定了基础
同时也为寻找鼻咽癌新的分子标志物提供了可能?
Abstract: 【Objective】 To analyze the status of DNA methylation in early nasopharyngeal carcinoma
and to find out the DNA methylation profile of early nasopharyngeal carcinoma preliminarily. 【Methods】 Four pairs of paraffin specimens of NPC(T1N0M0) and inflammatory nasopharyngeal tissues were collected to extracted DNA; The enriched genomic methylation fragments of two group specimens were used for high-throughput sequencing analysis by NimbleGen Human DNA Methylation 3 × 720K CpG Island Plus RefSeq Promoter Array. We scanned the hybridization signal
processed preliminary data and analyzed the genomic DNA methylation differences between early stage NPC and matched normal nasopharyngeal tissues. 【Results】 The number of methylated genes in all four cases of NPC paraffin specimens are 1134
and the number of methylated genes in the inflammatory nasopharyngeal tissues are 2260. Among the early stage NPC and compared specimens
the number of differentiation methylated genes are 218
and they might be the DNA methylation profiles of early stage NPC patients.In addition
the number of methylated genes in the four inflammatory nasopharyngeal tissues but not in the NPC paraffin specimens is 1558. 【Conclusion】 In this study
the DNA methylation profiles of early stage NPC were initially built. It laid a foundation for the further development of more precise profiles
meanwhile
it also provides possibility for looking for new molecular markers of NPC.
