Abstract: 【Objective】 To detect the methylation of ALX4 and SEPT9 in the peripheral blood samples from CRC patients and investigate multiplex methylight assay could be used for CRC detection as a non-invasive modality. 【Methods】 In the present study
we used a PCR multiplex method that in a fast and reliable manner to detect and quantify the methylation level of ALX4 and SEPT9 at the same time and applied this method to the detection of CRC through blood samples. 【Results】 In the peripheral blood plasma
the presence of ALX4
SEPT9 methylation was detected in 48% (24/50) and 75% (38/50) of our tested samples from CRC patients and 6% (3/50) and 4% (2/50) of normal controls
respectively. The difference in incidence of the methylation of CRC plasma samples compared with normal plasma samples was statistically significant (Pearson Chi-Square

the multiplex methylight assay for ALX4 and SEPT9 is an available high-throughput quantitative multigene methylation assay that can detect CRC using peripheral blood samples with a high sensitivity and specificity.
