摘 要: 【目的】 应用GoldeneyeTM 20A扩增体系,讨论IBS法和ITO法在全同胞鉴定中的应用价值。【方法】 选取150对全同胞和500对无关个体,用GoldeneyeTM20A体系进行19个常染色体STR基因座的扩增,计算状态一致性评分(IBS),用ITO法分别计算全同胞关系指数(FSI)、半同胞关系指数(HSI)及其比值(FSI:HSI)。【结果】 在检测GoldeneyeTM 20A体系19个基因座时,(1)当两个体的IBS≥22时,倾向于全同胞;当IBS≤13时,倾向于无关个体;当13<IBS<22时,无法给出倾向性意见。(2)当FSI<0.01时,倾向于无关个体;当FSI>1且FSI:HSI>1时,倾向于全同胞;当0.01≤FSI≤1或FSI>1而FSI:HSI≤1时,无法给出倾向性意见。【结论】 IBS 法和 ITO法都可以用在全同胞亲缘鉴定中,IBS法将作为新标准出台,其具有快速简便,高准确率的优势,而ITO法计算过程较为复杂,但具有较高的检出概率,是IBS法一种很好的补充。在实际检案中,可根据案件具体情况,综合运用。
Abstract:【Objective】 To discuss IBS and ITO methods in full sibling identificationby using GoldeneyeTM 20A PCR system. 【Methods】 A total of 150 pairs of full siblingsand 500 pairs of unrelated individuals were genotyped by GoldeneyeTM 20A system.State score (IBS), full sibling index (FSI), half sibling index (HSI) and the FSI:HSIratio were calculated. 【Results】 In GoldeneyeTM 20A system, (1) When two individuals’ IBS≥22, means it inclined to full sibling; When IBS of the two individuals≤13, inclinedto unrelated individuals; If 13<IBS < 22, means it cannot draw any conclusion. (2)If FSI>1 and FSI:HSI>1, means it inclined to full sibling; If FSI < 0.01, inclinedto unrelated individuals; If 0.01≤FSI≤1 or FSI>1, but FSI:HSI≤1, means it cannotdraw any conclusion. 【Conclusion】 Both IBS and ITO methods can be used in full siblingidentification. The IBS method has advantages of convenient, high speed, and highaccuracy. The ITO method relatively shows more complex, but it could be a supplementaryfor IBS. It suggests that in actual cases, according to the real situation, bothmethod could be used.
