摘 要: 【目的】 观察苯甲酸钠(SB)对PC12肾上腺嗜铬细胞瘤细胞凋亡的诱导作用及对酪氨酸羟化酶(TH)表达的影响。【方法】 以大鼠PC12细胞为多巴胺能神经元细胞模型,以10~ 50 mmol/L不同浓度的SB加入PC12细胞,培养24 h后,噻唑蓝(MTT)比色法检测细胞活性;DAPI染色观察细胞凋亡形态;流式细胞术检测细胞凋亡状况以及用Westernblot检测TH蛋白表达。【结果】 SB可呈浓度依赖性抑制PC12细胞活性,并诱导PC12细胞核形态呈凋亡改变;流式细胞术检测显示随着SB浓度增加,细胞凋亡率显著增加;低浓度(10mmol/L)SB作用下PC12细胞中TH表达增强,而高浓度(40 ~ 50 mmol/L)SB作用下TH表达减弱,同时,高浓度(40 mmol/L)SB作用下TH的表达减弱可呈时间依赖性。【结论】SB能够诱导PC12细胞凋亡,并伴随TH的表达改变。
Abstract: 【Objective】 To investigate the effects of sodium benzoate (SB)-induced apoptosis in adrenal pheochromocytoma (PC12) cell andthe expression of tyrosine hydroxylase (TH). 【Methods】 PC12 cells of rats were modelsfor dopaminergic neuron. MTT assay was used to examine the cell viability of PC12cells treated by different concentration (10-50 mmol/L)SB after 24 h. DAPI stain and flow cytometry were used to observe the morphologicalchanges of apoptosis and apoptotic rates respectively. The expression of TH weredetermined by Western blot.【Results】 SB could inhibit PC12 cells viability in dose-dependent manner. Apoptosis was obviously observed using DAPIstaining. Flow cytometry showed that apoptosis rates significantly increased withincreasing SB concentrations. The expression of TH in PC12 cells was increased whentreated with low dosage (10 mmol/L) of SB, whereas this expression was decreasedwhen treated with high dosage (40-50 mmol/L) of SB. Andthe expression of TH when treated with high dosage (40 mmol/L) of SB was decraesedin a time-dependent manner. 【Conclusion】 SB could induceapoptosis of PC12 cells and accompanied with change the expression of TH.
