摘 要: 【目的】 阐明BMP2是否参与了Decorin诱导的血管平滑肌细胞(VSMC)钙化。【方法】 本研究采用人血管平滑肌细胞钙化体外模型,用腺病毒载体过表达Decorin,检测骨相关蛋白BMP2、Msx2、Osterix的表达和细胞钙化程度。观察抑制或增加BMP2信号对Decorin诱导的血管平滑肌细胞钙化和Msx2、Osterix表达的影响。【结果】Decorin能促进人血管平滑肌细胞钙化,同时上调BMP2、Msx2、Osterix的表达水平。抑制BMP2能减轻Decorin诱导的细胞钙化,而增加BMP2加速Decorin诱导的细胞钙化。【结论】Decorin通过调节BMP2促进人血管平滑肌细胞钙化。
Abstract: 【Objective】 The study aims to elucidate whether Decorin regulatescalcification of vascular smooth muscle cells (VSMC) involving BMP2 signaling. 【Methods】In vitro model of human VSMC calcification was used in this study. Decorin was overexpressedin cultured human VSMC using adenovirus-mediated gene transfer.The expression of BMP2, Msx2 and Osterix was analyzed by qRT-PCRand calcification was assessed by alizarin red staining. In addition, we checkedthe role of BMP2 signaling in decorin-induced vascular calcificationby blocking or augmenting BMP2 signaling. 【Results】 We found that Decorin acceleratedhuman VSMC calcification and up-regulated the expressionlevel of BMP2、Msx2 and Osterix. Inhibition of BMP2 signaling attenuated Decorin-induced VSMC calcification, whereas augmentation of BMP2 signalingaccelerated Decorin-induced VSMC calcification. 【Conclusion】We demonstrate that Decorin induces human VSMC calcification via activation of BMP2signaling.
