【目的】 探讨激活核因子E2相关因子2(Nrf2)通路能否减轻高糖诱导足细胞的凋亡。【方法】体外培养小鼠足细胞,分为5组:对照组、高渗组、高糖组、高糖+叔丁基对苯二酚( tBHQ),高糖+N-乙酰半胱氨酸(NAC)。流式细胞仪检测细胞内活性氧和凋亡;Western blot 及real time- PCR检测Nrf2、醌氧化还原酶1(NQO1),血红素加氧酶(HO-1)的表达;检测足细胞丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)及过氧化氢酶(CAT)。【结果】 高糖组足细胞凋亡率、活性氧及细胞MDA高于对照组(P < 0.05),而该组足细胞SOD、GSH及CAT低于对照组(P < 0.05);高糖+NAC组的足细胞凋亡率、活性氧及细胞内丙二醛低于高糖组(P < 0.05);高糖+tBHQ组足细胞的总Nrf2及核Nrf2、 HO-1、NQO1的表达高于高糖组(P < 0.05),细胞内SOD、GSH及CAT高于高糖组(P < 0.05),高糖+tBHQ组足细胞的凋亡率、活性氧及细胞内丙二醛低于高糖组(P < 0.05)。【结论】 激活 Nrf2通路降低高糖诱导足细胞的氧化应激,减轻足细胞凋亡。
【Objective】 To investigate whether activation of nuclear factor erythroid-2 related factor (Nrf2) signal pathway reduced apoptosis in podocyte induced by high glucose. 【Methods】 Cultured mouse podocytes were divided into five groups: normal glucose (NG), mannitol group (MA), high glucose+ tert-Butyl-hydroquinone group (HG+tBHQ) and high glucose+N-acetylcysteine (HG+NAC). Reactive oxygen species (ROS) and apoptotic rate in podocytes were assessed by flow cytometry. The expression of Nrf2, quinone oxidoreductase-1(NQO1) and hemeoxygenase-1(HO-1) in podocytes were measured by Western blot and real time PCR, what’s more, the levels of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH) and catalase (CAT) in podocytes were measured at the same time. 【Results】 The apoptotic rate, ROS and MDA levels in podocytes exposed high glucose were higher than podocytes in NG group (P < 0.05), while the levels of SOD, GSH, and CAT in these podocytes were lower than podocytes in NG group (P < 0.05). The apoptotic rate, ROS and MDA levels of podocytes in HG+NAC group were lower than podocytes from HG group (P < 0.05). The expression of total Nrf2,nuclear Nrf2, NQO1 and HO-1 in podocytes from HG+tBHQ group were significantly increased than podocytes in HG group (P < 0.05), and the levels of SOD, GSH and CAT in podocytes from HG+tBHQ group were higher than podocytes in HG group (P < 0.05), what?蒺s more, the apoptotic rate, ROS and MDA levels in podocytes from HG+tBHQ group were lower than podocytes in HG group (P < 0.05). 【Conclusion】 Activation of Nrf2 signal pathway could alleviate oxidative stress and reduce apoptosis in podocytes induced by high glucose.
