摘要

【目的】探讨自噬在血管紧张素(Ang)Ⅱ诱导心肌细胞肥大中的作用.【方法】原代培养的乳鼠心肌细胞

采用Ang Ⅱ干预建立心肌肥大的模型;在光学显微镜下观察心肌细胞的形态

采用Real time PCR技术检测心肌细胞肥大标志物心房利钠多肽(ANP)的表达

Western blot法检测LC3蛋白的表达.【结果】 Ang Ⅱ可抑制自噬相关基因LC3蛋白的表达

自噬阻断剂3-甲基腺嘌呤(3MA)会促进AngⅡ诱导的心肌细胞肥大.【结论】 AngⅡ可能通过抑制自噬来诱导心肌细胞肥大.

Abstract

【Objective】 To investigate the role of autophagy in cardiac hypertrophy induced by angiotensin Ⅱ(Ang Ⅱ). 【Methods】Cardiac hypertrophy was induced by Ang Ⅱ incultured neonatal rat cardiomyocytes. Morphology and expression of hypertrophy marker atrial natriuretic peptide (ANP) were detected by optical microscope and real time-PCR. Autophagic marker

LC3 level was tested by Western blotting.【Results】AngⅡ increased volume of cardiomyocyte

and up regulated RNA expression of ANP. AngⅡ inhibited autophagy related gene LC3 protein expression. Blocking autophagy by 3-MA could significantly enhance myocardial hypertrophy induced by Ang II. 【Conclusion】 Ang Ⅱ may induce myocardial hypertrophy by inhibiting autophagy.
