MicroRNA-155 Inhibits Macrophagic Foam Cell Formation through Downregulating Expression of Scavenger Receptors[J]. Journal of Sun Yat-sen University (Medical Sciences), 2012, 33(2).DOI:
【Objective】 To study the effect of microRNA-155 (miR-155) on macrophagic foam cell formation. 【Methods】 Real-time RT-PCR was used to test the expression of microRNA-155. Western blot was used to determine the expression of scavenger receptors SR-A and CD36 in THP-1 macrophages. Receptor-specific binding and uptake of DiI-labeled ox-LDL (DiI-oxLDL) were examined by laser scanning confocal microscope. 【Results】 In THP-1 cells
80μg/mL oxidized low density lipoprotein (ox-LDL) induced up-regulation of miR-155 in a time-dependent manner. Ad-miR-155 transfection decreased the expression of SR-A and D36
and the uptake and binding of THP-1 cell with DiI-oxLDL. In contrast
miR-155 inhibitor increased the expression of SR-A and CD36
and the uptake and binding of THP-1 cell with DiI-oxLDL..【Conclusion】 MicroRNA-155 inhibits macrophagic foam cell formation through decreasing the expression of SR-A and CD36.
