【Objective】To explore Aβ25-35 -induced autophagy in HT22 cells and relationship with V-ATPase. 【Methods】 HT22 cell was divided into control group

A β treatment group
and A β + Bafilomycin A1 group. CCK8 assay was used to determine the cell vitality in different concentration of Aβ25-35 for 24 hours. LC3 puncta dots were observed by immune cell fluorescence. The expression of LC3 and V-ATPase were detected by Western blot analysis. 【Results】 Aβ25-35 in HT22 cells effected cell vitality in a time- and concentration-dependent manner. The amount of V-ATPase was correlated with the extent of autophagosome formation. Bafilomycin A1 could further induce V-ATPase and intracellular autophagosome accumulation(P ≤ 0.05). 【Conclusion】Aβ25-35 can induce the autophagy of HT22 cells and is relative with V-ATPase expression.
