【目的】 探讨沉默Pokemon基因对鼻咽癌细胞(CNE-2)放射敏感性的影响,寻求放射治疗鼻咽癌的新方法。 【方法】 首先利用siRNA预测软件得到3条siRNA序列,并制备合成siRNA片段,利用RT-PCR和免疫印迹法检测其沉默Pokemon基因的效果,筛选出沉默效果最佳的siRNA片段,转染CNE-2细胞后观察其对CNE-2细胞p53基因表达的影响、MTT法检测细胞的增殖活性、流式细胞术检测细胞的凋亡、克隆形成法检测转染siRNA联合放射处理后细胞的放射增敏比。【结果】 成功筛选出了沉默效应最佳的siRNA片段,转染CNE-2细胞后能显著降低其Pokemon基因的表达、提高p53基因的表达、降低细胞的增殖活性、加速细胞的凋亡,同时增加了CNE-2细胞的放射敏感性。【结论】 沉默Pokemon基因能增加鼻咽癌CNE-2细胞的放射敏感性,这为临床鼻咽癌的放射增敏治疗提供了新思路。
【Objective】 To investigate the effect of pokemon gene knockdown on radiation sensitivity of nasopharyngeal carcinoma cell (CNE-2). 【Methods】 Three siRNA sequences were designed through siRNA prediction software, and the best siRNA was screened by detecting the expression of Pokemon with RT-PCR and Western blotting. After the siRNA was transfected to CNE-2 cells, the p53 expression was investigated, and the cell proliferation was detected by MTT, and the apoptosis was detected by flow cytometry, and the radiosensitization ratio was detected by clone formation. 【Results】 The best siRNA was successfully screened. siRNA transfecting could decrease Pokemon expression, enhance p53 expression, reduce the cell proliferation, accelerate apoptosis, and augment the radiosensitization.【Conclusion】 Knockdown of pokemon could increase radiosensitization of nasopharyngeal carcinoma cell (CNE-2), which supplied a new idea of the nasopharyngeal carcinoma radiotherapy.
