Effect of Berberine on RLH Signaling Pathway in Lung of Mice Infected by Influenza A (FM1 strain)[J]. Journal of Sun Yat-sen University (Medical Sciences), 2016, 37(6).DOI:
【Objective】To determine the effect of berberine on RLH signaling pathway infected by influenza A (FM1 strain) in mice
and explore its anti-influenza mechanism. 【Methods】RIG-I?/? mice and normal wide-type C57BL/6 mice were intranasally inoculated with 50 μL LD50 influenza A (FM1 strain)
and treated with berberine and oseltamivir. The survival rates and pathological changes in the lung tissue after infection were examined. Using real-time fluorescence quantitative PCR reaction (RT-qPCR) and Western-blot to detect the RIG-I
NF-κB and MAVS expression of normal wide-type C57BL/6 mice.【Results】The berberine could protect the normal wide-type C57BL/6 mice from the viral infection and reduce the pathological degree of the viral pneumonia. In addition
the survival rate of berberine increased from 50% to 80% compared with virus group. RT-qPCR experiments and Western-blot experiments showed that berberine could reduce RIG-I
NF-κB and MAVS expression of virus-infected normal wide-type C57BL/6 mice. Compared with virus group
the expression of their mRNA reduced 42.28%
33.12%
and 48.12%
separately and the expression of protein reduced 17.37%
13.83%
and 28.91%
separately.【Conclusions】The berberine had good effect on prohibit influenza A virus as oseltamivir
which could regulate RLH signaling pathway of the infection.
