【目的】研究P-选择素对自发肠道腺瘤Apc Min/+ 小鼠中晚期肿瘤生长的作用。【方法】将P-选择素敲除小鼠与 Apc Min/+ 小鼠杂交,建立P-选择素敲除背景下的Apc Min/+ 小鼠模型(A M/+ Ps -/- 小鼠),将15周和24周的Apc Min/+ 小鼠和A M/+ Ps -/- 小鼠 的肠道亚甲蓝染色后,用倒置显微镜计数肠道肿瘤的数目并测量肿瘤的体积。【结果】在15周时,A M/+ Ps -/- 小鼠的肿瘤数目和 肿瘤体积与Apc Min/+ 小鼠相比没有明显差异。但在24周时,A M/+ Ps -/- 小鼠的肿瘤数目比Apc Min/+ 小鼠多,肿瘤体积比Apc Min/+ 小鼠 大,有显著统计学差异。而且A M/+ Ps -/- 小鼠的生存时间与Apc Min/+ 小鼠相比,也明显缩短,P = 0.0004。【结论】P-选择素的缺失 对Apc Min/+ 小鼠中期肿瘤生长没有明显影响,但是明显促进了Apc Min/+ 小鼠晚期肿瘤的生长。
【Objective】To investigate the effect of P-selectin on the intestinal tumorigenesis in the 15,24-week-old Apc Min/+ mice.【Methods】Male Apc Min/+ mice were mated with female P-selectin knockout(P-sel -/- )mice to generate A M/+ Ps -/- mice. The diameters and numbers of the intestinal tumors in the intestines of Apc Min/+ and A M/+ Ps -/- mice were measured using an inverted microscope. The tumor volumes were measured using the following equation:volume = 0.52 ×(length × width 2 ).【Results】There were no significant difference between the volume and number of intestinal tumor in 15-week-old Apc Min/+ mice and 15-week-old A+ P- mice. The volume and number of intestinal tumor were significantly increased in 24-week-old A+P- mice compared to 24-week- old ApcMin/+ mice. P-selectin deletion significantly prolonged the survival time of ApcMin/+ mice.【Conclusions】P-selectin deletion promotes the intestinal tumorigenesis in the 24-week-old ApcMin/+ mice.
