摘 要: 【目的】 检测未经过促排卵刺激的PCOS患者和正常排卵妇女卵巢组织中GDF9和BMP15的蛋白表达水平以及比较两组之间的表达差异。 【方法】 收集未经过卵巢刺激的PCOS患者(PCOS组)和正常排卵妇女(对照组)的卵巢组织,采用免疫组织化学染色的方法检测卵母细胞和颗粒细胞中GDF9和BMP15的蛋白表达水平。 【结果】 GDF9和BMP15蛋白在未刺激卵母细胞和颗粒细胞中均有表达,并且呈现阶段依赖性。GDF9蛋白在PCOS早期卵泡卵母细胞和颗粒细胞中的表达水平均低于对照组并且表达滞后(P < 0.05),BMP15蛋白在PCOS早期卵泡的颗粒细胞中也存在表达水平降低和滞后现象(P < 0.05),但是在PCOS卵母细胞中的表达与对照组没有差异(P > 0.05)。【结论】 GDF9和BMP15蛋白在未刺激卵母细胞和颗粒细胞中均呈现阶段依赖性表达;两种因子在PCOS早期卵泡阶段存在表达水平降低并且滞后现象,提示可能与PCOS卵泡发育紊乱有关。
Abstract:.【Objective】 To determine the expression levels of GDF9 and BMP15 in unstimulated ovarian tissues from PCOS patients and normal ovulatory women, and compare the differences between the two groups. 【Methods】 The unstimulated ovarian tissues were collected from PCOS patients and normal ovulatory women. Immunohistochemical staining was used to detect the expression levels of GDF9 and BMP15 both in oocytes and granulosa cells within the tissues. 【Results】 Both the GDF9 and BMP15 were stage dependently expressed in oocytes and granulosa cells within the ovarian tissues. The expression of GDF9 was lower and delayed in oocytes and granulosa cells of PCOS tissues than in control group (P < 0.05), the similar result was obtained for BMP15 in granulosa cell of PCOS tissues (P < 0.05). However, the expression of BMP15 was comparable in PCOS oocyte and normal control (P > 0.05). 【Conclusion】 The expression features of GDF9 and BMP15 demonstrate the stagedependent characteristic both in oocytes and granulosa cells without ovarian stimulation. The expression of two factors was reduced and delayed in the early follicular phase in PCOS tissues, suggesting that might be associated with aberrant PCOS follicular development.
