【目的】 探讨细胞因子信号转导负调控因子(SOCS3)诱导反复自然流产(RSA)患者蜕膜及绒毛组织Th1/Th2转换的作用。【方法】 采用ELISA方法检测27例RSA患者蜕膜及绒毛组织Th1细胞因子(IL-2、IFN-γ)和Th2细胞因子(IL-4、IL-10)的表达水平,采用流式细胞仪检测蜕膜及绒毛组织中Th1和Th2细胞水平及其比率,采用western-blot方法检测蜕膜及绒毛组织SOCS3蛋白表达水平,同时与30例正常妊娠行人工流产者比较。【结果】 RSA组患者蜕膜及绒毛组织中IL-2、IFN-γ表达水平明显高于对照组(P < 0.05),而RSA组患者蜕膜及绒毛组织中IL-4、IL-10表达水平明显低于正常对照组(P < 0.05);RSA组患者蜕膜及绒毛组织中Th1细胞水平及Th1/Th2比率明显高于对照组(P < 0.05),而Th2细胞水平明显低于对照组(P < 0.05);RSA组患者蜕膜及绒毛组织中SOCS3蛋白表达水平明显低于对照组(P < 0.05)。【结论】 RSA患者蜕膜及绒毛组织中SOCS3蛋白表达过低可诱导Th1/Th2失衡,使得Th1细胞介导的免疫损伤作用占主导地位,最终导致RSA发生。
【Objective】 To investigate the role of Th1/Th2 transform induced by suppressor of cytokine signaling (SOCS3) in decidua and villi of the patients with recurrent spontaneous abortion. 【Methods】 The expression levels of Th1 cytokines (IL-2, IFN- γ) and Th2 cytokines (IL-4, IL-10) in decidua and villus of RSA patients (27 cases) were detected by ELISA methods. The levels and ratio of Th1, Th2 cell in decidua and villus were detected by flow cytometry. The expression levels of SOCS3 protein in decidua and villi were detected by Western-blot, and 30 cases of normal pregnancy artificial abortion act as control group. 【Results】 The expression of IL-2, IFN-γ in decidua and villi of RSA group was significantly higher than that of the control group (P < 0.05), while the expression of IL-4, IL-10 in decidua and villi of RSA group was significantly lower than that of the control group(P < 0.05). The level of Th1 cell and ratio of Th1/Th2 in RSA group were significantly higher than that of control group (P < 0.05), while the level of Th2 cells was significantly lower than that of the control group (P < 0.05). The expression of SOCS3 protein in decidua and chorion of RSA group was significantly lower than that of the control group (P < 0.05). 【Conclusion】 The low expression of SOCS3 protein in decidua and villi of RSA patients could induce imbalance of Th1/Th2, immune injury of mediated by Th1 cell was dominate, eventually lead to the occurrence of RSA.
