CCK8检测细胞增殖情况;流式细胞术检测细胞凋亡;划痕试验和transwell检测细胞迁移能力;克隆形成试验检测细胞成瘤能力?【结果】 LMP2A基因转染的胃癌细胞生长速度增快?凋亡减少?迁移能力增强

在平板和软琼脂中集落形成能力增强?【结论】 LMP2A基因能明显增强胃癌细胞的恶性生物学行为?

Abstract

【Objective】 To investigate the effects of EBV LMP2A on biological characters of gastric cancer cells.【Methods】 Human gastric cancer cell line SGC7901 was transfected with a retroviral vector containing the LMP2A gene. The expression of LMP2A gene mRNA was detected by QRT-PCR. The cell proliferation was detected by CCK8 assay. Cell apoptosis were analyzed by flow cytometry. The migration of cell was measured with wound healing assay and Transwell test. Cell tumorigenicity was measured with colony formation assay.【Results】 The LMP2A gene-transfected gastric cancer cells grew vigorously and rapidly

had less apoptotic cells

formed more colonies and up-regulated the ability of migration. 【Conclusion】 The LMP2A gene can significantly enhance the malignancy of gastric cancer cells.
