【目的】 探讨MRI分子靶向示踪剂细胞间粘附分子链接微米级氧化铁颗粒(ICAM-MPIO)对放射性脑损伤早期无创性诊断的能力及敏感性。【方法】 制备分子靶向示踪剂ICAM-MPIO;RBI动物模型建立并分组(靶向组、非靶向组、对照组及钆增强组,每组6只);通过体外细胞及活体动物实验评价ICAM-MPIO对早期放射性脑损伤的靶向性,以及临床3.0T磁共振对ICAM-MPIO检测的敏感性。【结果】体外细胞实验显示ICAM-MPIO靶向识别激活内皮细胞,其结合效率约为非靶向组的5倍。活体动物实验显示ICAM-MPIO对早期放射性脑损伤具有明显的靶向作用,临床3.0T磁共振能清晰显示病灶区域ICAM-MPIO所引起的T2低信号;且靶向组T2信号(注射前/后:1310/944)减低幅度约为非靶向组(注射前/后:1 300/1 249)的7倍。【结论】 MRI分子靶向示踪剂ICAM-MPIO有望应用于早期无创性诊断放射性脑损伤。
Abstract: 【Objective】 To evaluate the ability and sensitivity of MRI targetedtracer ICAM-MPIO to diagnose the early stage of RBI. 【Methods】A new epitope-specific MRI tracer ICAM-MPIOwas compounded. Animal model of the early period of RBI were build and divided (targetedgroup, non-targeted group, control group, and Gd enhancedgroup, 6 rats per group). The targeted ability of ICAM-MPIObounded to early stage of RBI was evaluated in vitro and in vivo. And the sensitivityof clinical MRI was evaluated to discover ICAM-MPIO. 【Results】In vitro, ICAM-MPIO bound obviously to the irradiated EC.It was measured that the number of adherent microspheres on irradiated EC was 5fold than that of the non-targeted group. In vivo, ICAM-MPIO was targeted to the early stage of RBI, and clinical 3.0T MRI was sensitive to discover the negative signal intensity of ICAM-MPIO in the lesion. The decreased value of T2 signal intensityin targeted groups was 7-fold of non-targetedgroup. 【Conclusion】 This novel MR imaging tool ICAM-MPIOmay prove valuable for the early diagnosis of RBI non-invasively.
